En el RETÍCULO ENDOPLÁSMICO se une a oligosacáridos con unión N específica que se encuentran en las proteínas recientemente sintetizadas y funciona como CHAPERONA MOLECULAR que puede desempeñar un papel en el PLEGAMIENTO DE PROTEÍNAS o retención y degradación de proteínas mal plegadas. Además, la calreticulina es una forma de depósito principal para el CALCIO y funciona como molécula de señalización del calcio que puede regular la HOMEOSTASIS intracelular del calcio.
