Scope note: Estructuras anormales localizadas en varias partes del cerebro y compuestas de agrupaciones densas de filamentos helicoidales apareados (neurofilamentos y microtúbulos). Estas subunidades transversales dispuestas en pilas de doble hélice se tuercen hacia la izquierda, para formar filamentos acintados, que al parecer incorporan las siguientes proteínas: 1) los filamentos intermedios: neurofilamentos de alto a medio peso molecular; 2) las proteínas asociadas al microtúbulo: map-2 y tau; 3) actina; y 4) UBIQUITINAS. Como uno de los rasgos característicos de la ENFERMEDAD DE ALZHEIMER, los ovillos neurofibrilares eventualmente ocupan todo el citoplasma en ciertas clases de células en la neocorteza, el hipocampo, el tronco cerebral y el diencéfalo. El número de estos ovillos, tal como se ve en la histología postmortem, se correlaciona con el grado de demencia durante la vida. Algunos estudios sugieren que los antígenos de los ovillos se escapan hacia la circulación general tanto en el curso del envejecimiento normal como en los casos de la enfermedad de Alzheimer.

These double helical stacks of transverse subunits are twisted into left-handed ribbon-like filaments that likely incorporate the following proteins: (1) the intermediate filaments: medium- and high-molecular-weight neurofilaments; (2) the microtubule-associated proteins map-2 and tau; (3) actin; and (4) UBIQUITINS. As one of the hallmarks of ALZHEIMER DISEASE, the neurofibrillary tangles eventually occupy the whole of the cytoplasm in certain classes of cell in the neocortex, hippocampus, brain stem, and diencephalon. The number of these tangles, as seen in post mortem histology, correlates with the degree of dementia during life. Some studies suggest that tangle antigens leak into the systemic circulation both in the course of normal aging and in cases of Alzheimer disease.
