Nota de alcance: Mad2 es un componente del punto de control del ensamblaje del huso acromático. Se une e inhibe a la subunidad activadora del complejo promotor de la anafase, Cdc20, impidiendo el comienzo de la anafase hasta que todos los cromosomas están adecuadamente alineados en la placa ecuatorial. La Mad2 es necesaria para la captura apropiada de los microtúbulos en CINETOCOROS.
