In vivo, estos heterodúplex pueden producirse por mutación o recombinación genética; in vitro, se forman por hibridación de ácidos nucleicos. El análisis por microscopía electrónica de los heterodúplex resultantes facilita el mapeo de regiones de similitud (homología) de secuencias de bases de ácidos nucleicos.
