Scope note: Método in vitro para producir grandes cantidades de fragmentos específicos de ADN o ARN de longitud y secuencia definidas a partir de pequeñas cantidades de cortas secuencias flanqueadoras de oligonucleótidos (cebadores). Los pasos esenciales incluyen desnaturalización térmica de las moléculas diana de doble cadena, reasociación de los cebadores, iniciadores, con sus secuencias complementarias, y extensión de los cebadores reasociados mediante síntesis enzimática con la ADN polimerasa. La reacción es eficiente, específica y extremadamente sensible. Entre los usos de la reacción está el diagnóstico de enfermedades, detección de patógenos difíciles de aislar, análisis de mutaciones, pruebas genéticas, secuenciación del ADN y el análisis de relaciones evolutivas.

Scope note: In vitro method for producing large amounts of specific DNA or RNA fragments of defined length and sequence from small amounts of short oligonucleotide flanking sequences (primers). The essential steps include thermal denaturation of the double-stranded target molecules, annealing of the primers to their complementary sequences, and extension of the annealed primers by enzymatic synthesis with DNA polymerase. The reaction is efficient, specific, and extremely sensitive. Uses for the reaction include disease diagnosis, detection of difficult-to-isolate pathogens, mutation analysis, genetic testing, DNA sequencing, and analyzing evolutionary relationships.
