Scope note: Técnica de amplificación del ADN basada en el ligamiento de SONDAS DE OLIGONUCLEOTIDOS. Las sondas son diseñadas para el exacto emparejamiento de dos secuencias adyacentes de un ADN diana específico. La reacción en cadena es repetida en tres pasos en presencia de exceso de sonda: (1) desnaturalización por el calor del ADN bicatenario, (2) hibridación de las sondas con el ADN diana, y (3) unión de las sondas por la ADN ligasa termoestable. Después de repetir la reacción 20-30 ciclos se mide la producción de sonda ligada.

After the reaction is repeated for 20-30 cycles the production of ligated probe is measured.
