Scope note: Dibenzodiacepina tricíclica, clasificada como agente antipsicótico atípico. Se une a diversos tipos de receptores del sistema nervioso central y muestra un único perfil farmacológico. La clozapina es un antagonista de la serotonina, a la que se une fuertemente en los subtipos de receptores 5-HT 2A-2C. También muestra una marcada afinidad por algunos receptores dopaminérgicos, si bien muestra un débil antagonismo con el receptor D2 de dopamina, un receptor que se cree que modula la actividad neuroléptica. El principal efecto adverso asociado con la administracion de este agente es la agranulocitosis.

Clozapine is a serotonin antagonist, with strong binding to 5-HT 2A/2C receptor subtype. It also displays strong affinity to several dopaminergic receptors, but shows only weak antagonism at the dopamine D2 receptor, a receptor commonly thought to modulate neuroleptic activity. Agranulocytosis is a major adverse effect associated with administration of this agent.
