Scope note: Método de microscopia electrónica de transmisión analítica, en la que se usa un microscopio electrónico equipado con una lente de filtración de energía. El método se basa en el principio de que algunos ELECTRONES que pasan a través de la muestra pierden energía cuando ionizan la capa interna de electrones de los átomos de la muestra. La cuantía de la pérdida de energía depende del elemento. El análisis del espectro de la pérdida de energía (ESPECTROSCOPIA DE PÉRDIDA DE ENERGÍA DE LOS ELECTRONES) revela la composición elemental de la muestra. Se utiliza analítica y cuantitativamente para determinar cuales, en qué cuantía y dónde están los ELEMENTOS específicos de una muestra. Por ejemplo, se usa para la cartografía elemental del FÓSFORO para rastrear las cadenas de ÁCIDOS NUCLEICOS en complejos nucleoproteicos.

The amount of energy loss is dependent upon the element. Analysis of the energy loss spectrum (ELECTRON ENERGY-LOSS SPECTROSCOPY) reveals the elemental composition of a specimen. It is used analytically and quantitatively to determine which, how much of, and where specific ELEMENTS are in a sample. For example, it is used for elemental mapping of PHOSPHORUS to trace the strands of NUCLEIC ACIDS in nucleoprotein complexes.
