Scope note: Técnica que emplea un sistema instrumental para realizar, procesar y exhibir una o más mediciones de células individuales obtenidas de una suspensión celular. Las células generalmente se tiñen con uno o más colorantes fluorescentes específicos para los componentes celulares de interés, por ejemplo, el ADN, y la fluorescencia de cada célula se mide cuando atraviesa rápidamente el haz de excitación (láser o lámpara de arco de mercurio). La fluorescencia brinda una medición cuantitativa de diversas propiedades bioquímicas y biofísicas de la célula como base para diferenciación celular. Otros parámetros ópticos mensurables incluyen la absorción y la difusión de la luz, aplicándose esta última a la medición del tamaño, forma, densidad, granularidad de la célula y su absorción del colorante.

Cells are usually stained with one or more fluorescent dyes specific to cell components of interest, e.g., DNA, and fluorescence of each cell is measured as it rapidly transverses the excitation beam (laser or mercury arc lamp). Fluorescence provides a quantitative measure of various biochemical and biophysical properties of the cell, as well as a basis for cell sorting. Other measurable optical parameters include light absorption and light scattering, the latter being applicable to the measurement of cell size, shape, density, granularity, and stain uptake.
