Scope note: Sistemas enzimáticos que contienen una sola subunidad y requieren exclusivamente magnesio para su actividad endonucleolítica. Las correspondientes metilasas de modificación son enzimas separadas. Los sistemas reconocen secuencias específicas cortas del ADN y escinden en sitios que están en la secuencia de reconocimiento o cerca de ella, produciendo fragmentos bicatenarios específicos con 5'-fostafos terminales. Las enzimas de diferentes microorganismos con la misma especificidad se denominan isoesquizómeros. EC 3.1.21.4.
