Scope note: Una dNTPasa y trifosforilhidrolasa de restricción del hospedador que contiene un MOTIVO ESTÉRIL ALFA aminoterminal y un dominio central conservado de ASPARTATO e HISTIDINA (HD). Actúa sobre el ARN monocatenario dando lugar a desoxinucleósidos y trifosfato y funciona como defensa antiviral mediante su actividad dNTPasa que reduce la concentración intracelular de dNTP por debajo de la requerida para la transcripción inversa retroviral en las CÉLULAS DENDRÍTICAS y CÉLULAS MIELOIDES. También tiene actividad RIBONUCLEASA que bloquea la replicación precoz de los retrovirus como el VIH-1. Las mutaciones del gen SAMHD1 se asocian con el síndrome de Aicardi-Goutieres tipo 5 y con el LUPUS asociado con eritema pernio (chilblain) tipo 2 (CHBL2).

Scope note: A host restriction triphosphorylhydrolase and dNTPase that contains an N-terminal STERILE ALPHA MOTIF and central, conserved ASPARTATE and HISTIDINE (HD) domain. It acts on single-stranded RNA, yielding deoxynucleosides and triphosphate, and functions in anti-viral defense through its dNTPase activity, reducing cellular dNTP levels below what is required for retroviral reverse transcription in DENDRITIC CELLS and MYELOID CELLS. It also has RIBONUCLEASE activity which blocks early replication of retroviruses such as HIV-1. Mutations in the SAMHD1 gene are associated with type 5 Aicardi-Goutieres syndrome (AGS5) and type 2 chilblain LUPUS (CHBL2).
