Scope note: Cepas de ratones cuyo genoma contiene un gen en el que su función ha sido alterada, o bloqueada. Un método común de producir genes alterados utilizando la tecnología del ADN RECOMBINAdo es alterar la secuencia normal del ADN del gen en estudio para impedir la síntesis del producto normal del gen. Las células clonadas en las que esta alteración se produce con éxito se inyectan en los EMBRIONES de ratones, obteniéndose ratones quiméricos. Estos ratones se aparean para producir una cepa en la que todas las células contienen el gen alterado. Los ratones knockout se utilizan como MODELOS ANIMALES EXPERIMENTALES para diversas enfermedades (MODELOS ANIMALES DE ENFERMEDADES) y para esclarecer las funciones de los genes.

Scope note: Strains of mice in which certain GENES of their GENOMES have been disrupted, or "knocked-out". To produce knockouts, using RECOMBINANT DNA technology, the normal DNA sequence of the gene being studied is altered to prevent synthesis of a normal gene product. Cloned cells in which this DNA alteration is successful are then injected into mouse EMBRYOS to produce chimeric mice. The chimeric mice are then bred to yield a strain in which all the cells of the mouse contain the disrupted gene. Knockout mice are used as EXPERIMENTAL ANIMAL MODELS for diseases (DISEASE MODELS, ANIMAL) and to clarify the functions of the genes.
