Metila la LISINA 9 (H3K9me) y la LISINA 27 (H3K27me) de la HISTONA H3, lo que ocasiona la represión transcripcional del gen diana afectado. EZH2 también metila proteínas no histónicas tales como el FACTOR DE TRANSCRIPCIÓN GATA4 y el receptor nuclear RORA. Regula los RELOJES CIRCADIANOS mediante la metilación de histonas de las REGIONES PROMOTORAS de los genes de proteínas circadianas, y su actividad represora también es importante para la identidad y diferenciación de las CÉLULAS MADRE EMBRIONARIAS.
