Scope note: Ubiquitina-ligasas E3 que constan de cuatro DOMINIOS WW. Mediante sus dominios HECT del extremo C, captan la UBIQUITINA, en forma de tioéster, de las ENZIMAS DE CONJUGACIÓN DE LA UBIQUITINA E2 y la transfieren específicamente al residuo de LISINA número 63 (Lys-63) de las proteínas sobre las que actúan. Las dianas NEDD4 incluyen muchas proteínas y receptores con importantes funciones para el crecimiento celular y la homeostasis, como los receptores VEGFR-2, el RECEPTOR TIPO 1 DE FACTOR DE CRECIMIENTO DE FIBROBLASTOS y el RECEPTOR ERBB-4. Juegan un papel crítico en la internalización de estos receptores y en su degradación por los LISOSOMAS; así mismo, también forman parte del complejo ESCRT en la LIBERACIÓN DE VIRUS.

NEDD4 targets include many proteins and receptors with important functions for cell growth and homeostasis such as VEGFR-2; FGFR1 TYROSINE KINASE; and ERBB-4 RECEPTOR. They play a critical role in the internalization of these receptors, their degradation by LYSOSOMES, and also function as part of the ESCRT complex in VIRUS RELEASE.
