Scope note: Glicoproteínas precursoras ricas en cisteína, codificadas por el gen de las granulinas (GRN). Contienen repeticiones de granulinas separadas por secuencias de unión que son escindidas por diversas proteasas para liberar los PÉPTIDOS DE GRANULINA individuales. La progranulina de mamíferos está compuesta de una repetición de media granulina seguida de siete repeticiones de granulina. Las GRANULINAS de mamíferos se denominan PARAGRANULINA y granulina 1-7 por su orden en el propéptido de progranulina. La progranulina tiene efectos similares a los de los factores de crecimiento y se expresa en una amplia variedad de tejidos. La infraexpresión de progranulina se asocia a enfermedades neurodegenerativas tales como la DEMENCIA FRONTOTEMPORAL y la LIPOFUSCINOSIS CEROIDE NEURONAL, mientras que la sobreexpresión se asocia a enfermedades metabólicas tales como la OBESIDAD y la RESISTENCIA A LA INSULINA.

They contain granulin-repeats separated by linker sequences that are cleaved by a variety of proteases to release the individual GRANULIN PEPTIDES. The mammalian progranulin is composed of a half-granulin repeat followed by 7 granulin repeats. The mammalian GRANULINS are named PARAGRANULIN and granulin 1-7 for their order in the progranulin propeptide. Progranulin has growth-factor like effects and is expressed over a broad range of tissues. Under-expression of progranulin is linked to neurodegenerative diseases including FRONTOTEMPORAL DEMENTIA and NEURONAL CEROID LIPOFUSCINOSIS, while over-expression is linked to metabolic diseases including OBESITY, and INSULIN RESISTANCE.
